The investigation into Hill's death quickly focused on a person of interest, leading law enforcement to a residence
in North Topeka. A tense standoff ensued, with the Topeka Police Department's Response Team and crisis negotiators engaging with an individual who had barricaded himself inside the home. After several hours, authorities successfully took 47-year-old Charles Melvin Wilson II into custody in the early morning hours of the following day.
Wilson was subsequently charged with first-degree murder in the death of Lisa Hill, along with criminal possession of a firearm by a convicted felon and possession of methamphetamine. His bond was set at one million dollars. The case against him was built on evidence gathered at the scene and through the course of the investigation, which pointed to him as the perpetrator of the fatal shooting.

Just 13 days prior to her murder, on April 24, 2020, her 26-year-old son, Joseph Ross Hill, was also shot and killed in Topeka. The investigation into Joseph's death was ongoing when his mother was killed, and Charles Melvin Wilson II was identified as a person of interest in that case as well.
